Pregnenolone, is a steroid hormone which is synthesized in our body from cholesterol and serves as a precursor progestogens, mineralocorticoids, glucocorticoids, androgens, and estrogens, as well as the neuroactive steroids.
Although pregnenolone is considered a prohormone, it has various biological effects of its own: 1) it behaves as a neuroactive steroid and improves memory and cognitive function; 2) it enhances myelinization of neurons; 3) it protects brain from the toxic effect of cannabis ant its metabolites; 4) it regulates activity of specific neurotransmitter-gated channels such as NMDA and GABA(A), and therefore controls glutamate-NO-cGMP pathway; 5) it possesses anti-inflammatory activity and reduces inflammation in skin and colon; 6) it indirectly controls calcium entry in liver cells (hepatocytes) and in pancreatic islands and triggers insulin release; 7) it regulates production of hyaluronic acid by synovial fibroblasts (joint cavity lining cells).
Known clinical applications of pregnenolone include treatment of adrenal insufficiency, treatment of anxiety, treatment of schizophrenia, treatment of psoriasis and rheumatoid arthritis.
